Scope
Scope: This standard specifies the relevant terms and definitions for the production of mushroom cultivation seeds using liquid strains, the preparation of liquid strains and related technical requirements, the process of cultivation seed production, quality inspection of cultivation seeds, production records and archiving, sample retention, and labeling and packaging. This standard applies to the industrialized production of mushroom cultivation seeds. Main technical content: 1. Production process: - Master strain shake flask - Liquid strain fermentation tank - Liquid strain culture medium preparation - Bagging and sterilization - Cooling and inoculation - Cultivation seed medium preparation - Master strain medium preparation - Use of potato-glucose agar medium (PDA) specified in Appendix A.1 or comprehensive potato-glucose agar medium (CPDA) specified in Appendix A.2. - Liquid strain culture medium formulations should be appropriate for the mushroom variety and local raw materials, with strict control of initial pH. - Cultivation seed medium formulations should refer to the NY/T 528 standard, Appendix B - Common cultivation medium formulations and their applicable varieties, and select according to the mushroom variety and local raw materials. - After subculturing the master strain, samples shall be sampled and subjected to sensory inspection and mycelial microscopic characteristics testing according to the NY/T 1846 standard. Qualified strains shall enter the production process. - The quality requirements for master strains shall comply with the NY/T 1742 standard on master strain production. 2. Shake flask liquid strain production: - Media dispensing and sterilization: - The shake flask liquid strain medium shall be dispensed into containers at a volume of 1/4 to 1/3 of the container, and immediately sterilized after dispensing. - Sterilization shall be carried out in a conv
