Scope
Scope: This document describes the methods for sampling, isolating, and archiving pine wood nematodes. This document is applicable to the sampling and isolation of pine wood nematodes in pine plants and products in epidemic areas. Main technical content: This operating procedure standardizes the types of samples, sampling tools, and operating times for sampling pine wood nematodes. Based on previous experience and practical work, it summarizes a complete set of sampling methods and isolation techniques. The specific techniques are as follows: (1) Conduct disease grading for infected pine trees and determine the sampling sites based on the disease grade; (2) Take the small plot as the unit and determine the amount of sampling and testing based on the number of diseased and dead pine trees in the small plot; (3) For sawn timber samples, consider two situations: round logs and other semi-finished wood products; (4) Pack the collected samples in plastic bags and mark them with a marker pen, then store them at 4℃ under humid conditions; (5) Manage the sample information in an archive; (6) The isolation methods for different types of samples are different. For branches, wooden strips, and wood chips, choose the Bellman funnel method; for discs, choose either the negative pressure separation method or the Bellman funnel method.
