This document is applicable to the production of tissue culture seedlings of bulbous yellow yam. Main technical content: 1. Preparation of culture medium mother solution: 2.1 Preparation method and storage of mother solution: The mother solution is prepared in strict accordance with the methods specified in GB/T 603, and the water used meets the specifications and test methods for analytical laboratory water in GB/T 6682. The commonly used reagents are prepared into a mother solution with a concentration 10-100 times higher than that required by the culture medium, and the mother solution is stored in a refrigerator at 4°C. 2.2 Preparation of macronutrient mother solution: Weigh 33 g of ammonium nitrate, 38 g of potassium nitrate, 3.4 g of potassium dihydrogen phosphate, 7.4 g of magnesium sulfate heptahydrate, and 8.8 g of calcium chloride dihydrate, respectively. Add a small amount of distilled water to each container and dissolve them thoroughly with a glass rod. Then, adjust the volume to 1000 ml in a volumetric flask and store them in a brown wide-mouth bottle with a label. 2.3 Preparation of micronutrient mother solution: Weigh 0.166 g of potassium iodide, 1.24 g of boric acid, 3.38 g of manganese sulfate tetrahydrate, 1.72 g of zinc sulfate heptahydrate, 0.05 g of sodium molybdate, 0.005 g of copper sulfate pentahydrate, and 0.005 g of cobalt chloride hexahydrate, respectively. Mix them in a beaker with a small amount of distilled water and dissolve them thoroughly. Then, adjust the pH to 5.5 and add water to 1000 ml in a volumetric flask. St
