Scope

Scope: This document specifies the technical content of equipment, reagents, and planning in the process of tissue culture and propagation of apple (Malus spp.) rootstocks; preparation of culture media; disinfection and sterilization of the tissue culture room; production of tissue culture seedlings of apple rootstocks; quality and packaging of tissue culture seedlings; labeling and transportation, etc. This document is applicable to apple rootstocks (JM series, CG series, M series, and Qingzhen series) for tissue culture and propagation. Main technical content: For the JM series, LS culture medium is used; for the CG series, QL culture medium is used; for the M series and Qingzhen series, MS culture medium is used, with the addition of suitable concentrations of various growth hormones. Plants with stable traits, robust growth, no pests or mechanical damage are selected from the mother plant nursery. In spring, young shoots with several small leaves emerging from leaf buds or unlignified or semi-lignified shoots are collected. Leaves that have unfolded on the shoots should be removed, but small unfolded leaves can be left unremoved. One-year-old branches can also be selected, and the bases are soaked in water indoors for germination. After the leaf buds sprout and grow into young shoots, they should be collected promptly. After sterile operation, the explants are subjected to primary and secondary propagation and rooting culture (the culture medium formula is detailed in Annex 3). The inoculation materials for the primary and secondary propagation and rooting culture are placed in the incubator or culture room. The culture conditions are: temperature (24±1)°C, light intensity 1500Lx-2000Lx, 10h-12h of illumination and 10h-12h of darkness. After 8-12 generations of propagation (determined by different rootstock varieties), the propagation coefficient can reach about 5.82, and robust young shoots with a height of more than 2cm can be cut and transferred to rooting
