Scope
Scope: This document establishes the process for the detoxification and seedling cultivation of sweet potatoes in Suixi County, Zhanjiang City, and specifies the operational instructions and conversion conditions for detoxification tissue culture seedling breeding, greenhouse cultivation, large-scale field propagation of detoxified seedlings, seedling harvesting, and seedling transportation, as well as describing the content of file management. This document is applicable to the production of detoxified sweet potato seedlings within the Suixi County area of Zhanjiang City. Main technical content: The seedling cultivation process consists of five stages: detoxification tissue culture seedling breeding, greenhouse cultivation, large-scale field propagation of detoxified seedlings, seedling harvesting, and seedling transportation. 5. Detoxification tissue culture seedling breeding 5.1 Variety selection: Select sweet potato varieties that have passed variety authentication (identification) or registration, and are suitable for local cultivation. Select tubers or seedlings with typical variety characteristics, strong stem and leaf growth, no leaf spots, mosaic, curling, shriveling, or yellowing. 5.2 Pre-disinfection: 5.2.1 Laboratory disinfection: Use potassium permanganate and formaldehyde to disinfect the laboratory space 7 days in advance. First, place potassium permanganate in a container, then add formaldehyde solution and fumigate for 24 hours. Ventilate until the laboratory is odorless. The dosage is 20 mL of 40% formaldehyde and 15 g of potassium permanganate per 1 m3 of space. 5.2.2 Instrument disinfection: Use ultraviolet lamps to irradiate the ultra-clean workbench for 30 minutes, and disinfect the workbench surface and inner walls with 75% alcohol.
