Scope
Scope: This document specifies the preparation of dilution protection solution, dilution methods, dilution ratios, and cooling and equilibration for semen dilution technology. This document is applicable to semen dilution. Main technical content: 3. Terms and definitions. There are no terms or definitions that need to be defined in this document. 4. Preparation of dilution protection solution. Requirements for the preparation of dilution protection solution for frozen semen should use double-distilled water, fresh egg yolk, and chemically pure reagents, etc. All equipment used must be clean, dry, and sterile. All dilution solutions (except egg yolk) must be filtered and boiled and sterilized in a water bath, cooled to 35°C–38°C, and then added with egg yolk and antibiotics. Glycerol should be sterilized and disinfected in a water bath for 30–45 minutes. The prepared dilution solution should be labeled and the stoppers of the triangular flasks sealed with silicone rubber before being stored in a 4.0°C refrigerator. The dilution protection agent must be prepared and used immediately. All equipment used for preparation should be clean and disinfected. The chemical reagents used (of chemical purity or higher) include trisodium citrate, fructose, sucrose, lactose, and glycerol. Fresh eggs from healthy chicken farms should be used for egg yolk. Double-distilled water should be used for all dilution solutions. The dilution solution should be operated according to the following regulations: a) Base solution: 1.5 g of sodium citrate, 3.0 g of glucose, 5.0 g of lactose, and adding distilled water to 100.0 mL, and sterilizing for use. b) Dilution solution: Adding 100,000 units of penicillin and streptomycin to the disinfected base solution (75.0 mL), 20.0 mL of egg yolk, and 5.0 mL of glycerol. c) Adding 100,000 units of penicillin and streptomycin to each 100.0 mL of all dilution solutions immediately before use. 5.
