Scope

Scope: This protocol specifies the technical terms and definitions of tissue culture for the geographical indication product "Shibing Prince Parrotia", including tissue culture, acclimatization, and propagation. This protocol applies to the production and propagation of detoxified tissue culture seedlings of the geographical indication product "Shibing Prince Parrotia". Main technical content: 1. Tissue culture 1.1 The requirements for the tissue culture room shall comply with GB 19489 and GB/T 22278. 1.2 The culture medium shall be based on MS medium, and the reagents used shall comply with the provisions of GB 437, GB/T 628, GB/T 647, GB/T 659, GB/T 664, GB/T 666, GB/T 671, GB/T 1272, GB/T 1274, GB/T 1401, and GB/T 15899. 1.3 Preparation and storage of culture medium stock solution: Prepare the culture medium formula several times larger, typically a 10-100-fold concentrated solution. Reagents shall be prepared in accordance with the methods specified in GB/T 603, and the water used shall comply with the requirements of GB/T 6682. The drug spoon used for weighing reagents must be dedicated for specific drugs. Label the prepared stock solution with the name of the solution, the preparation multiplier, and the date, and store it in a refrigerator at 2℃~4℃ for no more than 90 days. If mold or precipitation occurs during storage, the solution shall be re-prepared. Preparation and storage of plant growth regulators: Prepare various plant growth regulators separately with a concentration of 50mg/100mL~100mg/100mL. Most plant growth regulators are difficult to dissolve in water. The dissolution methods are as follows: NAA, IBA, and IAA are usually dissolved in a small amount of 95% ethanol, then heated to dissolve, and finally diluted with distilled water.
