Scope
Scope: This standard specifies the basic requirements for the production of black fungus liquid strains, including production sites, plant layouts, facilities and equipment, production processes, quality requirements, technical requirements, production records and archiving, and strain storage. This standard applies to the production of black fungus liquid strains; Main technical content: 1. Production process: - Mother strain preparation: Shake flask liquid strain cultivation medium preparation - Material loading: Material loading and sterilization - Cooling: Cooling after sterilization - Inoculation: Inoculation into fermentation tanks - Liquid strain cultivation medium: Use the potato glucose agar medium (PDA) specified in Appendix A.1 or the comprehensive potato glucose agar medium (CPDA) specified in Appendix A.2. For liquid strain cultivation medium, use an appropriate formula according to the requirements of black fungus varieties and local raw materials (see Appendix B) and strictly control the initial pH. - Mother strain: After secondary culture of the mother strain, samples shall be sampled and subjected to sensory inspection and mycelial microscopic characteristics inspection according to the provisions of NY/T 1846. Qualified strains shall enter the production process. The quality requirements of the mother strain shall comply with the provisions of GB 19169 and NY/T 1742 regarding the quality of the mother strain. - Shake flask liquid strain production: - Medium filling and sterilization: The filling volume of the shake flask liquid strain cultivation medium shall be 1/4 to 1/3 of the container volume. Immediately sterilize after filling. According to the routine high-pressure steam sterilizer operating procedures, maintain a temperature of 0.1Mpa to 0.12Mpa (121°C to 126°C) for 25 minutes. After sterilization, the shake flask liquid strain cultivation medium shall be cultured at 30°C ± 1°C for 48 hours. Obs
