Scope

Main Technical Content: 4. Basic Conditions - Technicians, site selection, plant layout, and equipment facilities shall comply with the requirements of NY/T 528. - Liquid strains shall have an independent cultivation room, an air purification system, and temperature control facilities, equipped with instruments and equipment necessary for liquid strain production, cultivation, and testing. 5. Seed Source - Seed varieties shall be selected from those approved by the national or provincial authorities; or varieties with clear genetic characteristics that have undergone two or more production cycle trials and are suitable for facility cultivation and industrialized farming. 6. Solid Strains - Mother Strains 6.1.1 Containers: Use 18 mm × 180 mm or 20 mm × 200 mm glass test tubes. 6.1.2 Culture Medium: Potato-Glucose Agar (PDA): 200 g potato, 20 g glucose, 20 g agar, and 1000 mL water. 6.1.3 Sterilization Preparation: After filtering the boiled potato juice, add the remaining reagents and water to 1000 mL. Heat until the agar melts and mix well. Distribute into test tubes, with the amount of culture medium occupying 1/5 to 1/4 of the tube capacity. Sterilize at 121°C for 30 minutes. 6.1.4 Cooling: After sterilization, cool the culture medium to 65°C ± 5°C. Place it on a slant in a clean environment, with the slant length not exceeding 2/3 of the tube length. After the slant solidifies, test the sterilization effect of the test tubes according to NY/T 528. 6.1.5 Inoculation and Cultivation: Carry out in accordance with NY/T 528 and make appropriate markings. After inoculation, place the test tubes in a 23°C–25°C environment without light exposure until the mycelium covers more than 2/3 of the slant.
