Main Technical Content: 5. Technical Requirements The inoculation must be performed under aseptic conditions to avoid contamination by impurities. During the cultivation of the fungal strain, regularly check the temperature and humidity of the incubator where the culture is grown; inspect the cotton plugs, lids, and culture medium of the incubation vessels for signs of mold growth. If abnormalities are detected, immediately remove the tube and repurify it. After culturing, replenish any empty tubes. 6. Methods and Steps Fungal strains are typically divided into three levels: 1. First-level strains, also known as mother strains or test tube strains. They can be obtained using tissue isolation or spore isolation methods. 2. Second-level strains, also known as original strains, are produced by inoculating the mycelium of the mother strain into sawdust culture medium. 3. Third-level strains, also known as cultivation strains, are produced by further propagating the original strains. These strains are directly used in production and are therefore also called production strains. After continuous propagation of the mother strain to the original strain and cultivation strain, the number and strength of the mycelium increase, enabling excellent fruiting bodies to be grown using this mycelium in production. 6.1.1 Preparation of Mother Strain Culture Medium Prepare the mother strain culture medium using PDA-enriched medium: - 200g potatoes - 20g glucose - 20g agar - 1.5–2g KH2PO4 - 0.5–1g MgSO4 - 10g peptone - 1000mL water Preparation method: - Peel and remove the bud eyes from 200g potatoes, cut them into small strips, and place them in an aluminum pot with 1000mL water. Add 20g cotton seed husks and boil for 20–30 minutes until the potatoes are soft but not mushy. Filter the solution through a 6–8-layer gauze, add 20g glucose, 1.5–2g KH2PO4, 0.5–1g MgSO4, and 10g peptone to make up to 1000mL, melt the agar, and dispense into test tubes (
