Scope

This document describes the experimental method for determining the intrinsic clearance rate of chemicals in vitro using the S9 subcellular fraction of rainbow trout liver, including the experimental principle, information on the test substance, reference substances, experimental preparation, experimental procedure, data processing, quality assurance and quality control, and result reporting.
This document is suitable for determining the intrinsic clearance rate of chemicals in vitro, and the minimum in vitro first-order clearance rate constant (Ke) can be accurately quantified as approximately 0.05 h-1 to 0.14 h-1. If necessary, it can also be used to identify metabolites.
